| Description: | PDE4 catalytic activity was monitored by measuring the hydrolysis of [3H]-cAMP to [3H]-AMP using PDE-SPA kit (Amersham International). [3H]-AMP was captured by the SPA beads, and quantified on a Wallac-Microbeta scintillation counter. A linear reaction progress curve (30 min) and a linear enzyme concentration response (0 to 500 pM) were observed. The IC50 value was calculated from an 11-point dose-response curve performed in duplicate using a 4-parameter least-squares nonlinear regression fit. |
