| Description: | The reaction mixture consisted of FAS thioesterase domain (FASTE) in buffer, which was preincubated with test compounds for 30 min. The reaction was initiated by addition of substrate 4-MUH. The resulting fluorescence from liberated 4-methylumbelliferone was measured every 5 min to generate concentration-response curves from which IC50 values were determined. The inhibition constants (Ki) were calculated using the equation of Cheng and Prusoff. |
