| Description: | The reaction mixture contained 1.5 mL of 50 mM potassium phosphate buffer (pH 7.5), 1 mL test sample solutions (5, 10, 25, 50, and 100 µM) was dissolved in DMSO, 0.5 mL of XO enzyme solution (0.2 units/mL of xanthine oxidase in phosphate buffer). The reaction mixture was pre-incubated at room temperature for 15 min, followed by the addition of 1 mL of substrate solution (0.10 mM of xanthine). The mixture was further incubated at room temperature for 30 min and stopped with the addition of 1 mL of 1 M HCl. Allopurinol was used as a positive control and the inhibitory effect of XO was measured spectrophotometrically at 292 nm under aerobic condition. |
