| Description: | NanoLuc enzyme was diluted to 0.4 ng/mL in CO2 independent media + 10% FBS to make the detection reagent. A 3× dilution series of each inhibitor was then made in the detection reagent. A "no inhibitor" control was also made for each sample. A total of 50 μL of each inhibitor dilution was mixed with 50 μL of NanoGlo buffer containing 20 μM furimazine (final furimazine concentration is 10 μM, which is atKM), and luminescence was measured. Each sample was normalized to the "no inhibitor" control. |
