Comments: | Assay Overview: The purpose of this assay is to determine dose-response curves for 20 compounds that demonstrated differential activities in a set of experiments entitled, "Fluorescence polarization-based confirmation biochemical high throughput screening assay to identify inhibitors of tRNA 2'-phosphotransferase (TPT1)" (AID 2149) and "Fluorescence polarization-based counterscreen assay for inhibitors of tRNA 2'-phosphotransferase (TPT1): biochemical high throughput screening assay to identify inhibitors of RNAse T1" (AID 2153). Compounds with a greater activity in the TPT1 assay compared to the RNase T1 counterscreen were selected and sorted by their distance (D) from a state where percent inhibition in both assays were equal according to the following equation: D = square root ( squared ( "% inhibition TPT1 assay" - "% inhibition RNase T1 counterscreen" ) / 2 ) The 20 compounds with the highest D value were selected for dose-response determinations.
