| Description: | The compound inhibitory activity was determined by incubation with purified recombinant Chk1 and biotinylated peptide substrate in the presence ATP/[gamma-33P] ATP. After incubation, the reaction was stopped and the phosphorylated substrate was separated from nonincorporated radioactive ATP using SPA beads. Incorporated radioactivity was measured using TopCount NXT microplate scintillation counter. IC50 values were determined using XLFit within Activity Base. |
