| Description: | Inhibition constants were calculated by assessment of the reduction in the formation of o-nitrophenol, as monitored by a spectrophotometric assay at 420 nm. The data was recorded at 15 s intervals for 5 min. All reactions were performed in duplicate, and at least 8 concentrations of inhibitor were used to determine Ki. Examination of curve fits to identify the best model for enzyme inhibition. Ki values were calculated from sigmoid curve fits using GraphPad Prism to be the best fit for the experimental data. |
