Assay Name:  Protease Inhibition Assay
Description:  IC50 values are determined by inhibition of the cleavage of the peptidic substrate [V-S-Q-N-(beta-naphthylalanine)-P-I-V]. The IC50 value represents the concentration of inhibitor required to inhibit cleavage of substrate by 50%. The assay is performed by addition of purified enzyme to a mixture of inhibitor and the above substrate at pH 5.5. The reaction medium is incubated for 30 min at 37 °C followed by quenching with H3-PO4 and then analyzed by reverse phase HPLC UV (220 nm) detection.
