| Description: | Inhibitory activities of NAPAs were measured by fluorescence anisotropy competition experiments. Protein was incubated with fluorescein-labeled human p53 peptide in the presence of test compound. Experiments were performed with a 70-fold molar excess of protein over labeled p53-peptide to ensure that the peptide is completely bound by the protein. Under these conditions, there is no difference in the minimum fluorescence polarization values between direct and competition assays. Fluorescence anisotropy was measured at 25 deg C on a Beacon 2000 (PanVera, Madison, WI). Data points were measured in triplicate. IC50 values were calculated by nonlinear regression curve fitting. |
