| Description: | The test compounds and enzyme were mixed in buffer, and the substrate and [gamma-33P] ATP/ATP were added to the mixture to initiate the reaction. After allowing the reaction to proceed at room temperature for 120 min, wheat germ agglutinin-coated SPA beads (Amersham) was added. The mixture was left to stand for 5 min and then centrifuged. Radioactivity was measured using TopCount (Packard). IC50 values represent means of at least two separate determinations with typical variations of less than +/- 20%. |
