| Description: | Assays were performedby incubating SENPs with various concentrations of the inhibitor (0−60 μM) at RT for 10 min in assay buffer (50 mM Tris, pH 7.4, 100mM NaCl, and 10 mM DTT). SENP concentrations were 32−50 nMwhen 50 μg/mL of the final substrate YFP-SUMO-ECFP (YSE) fusionprotein was added. The mixture was incubated (37 °C, 15 min),followed by SDS-PAGE and Coomassie staining for visualization. |
