| Description: | Briefly, 1,3,4-thiadiazole derivatives were tested for diphenolase inhibitory activity of tyrosinase using L-DOPA (dihydroxyphenylalanine) as substrate. All the compounds were dissolved in DMSO and its final concentration in the reaction mixture was 2.0%. Thirty units of mushroom tyrosinase (0.5 mg/ml) were first pre-incubated with the compounds, in 50 mM phosphate buffer (pH 6.8), for 10 min at 25 °C. Then the L-DOPA (0.5 mM) was added to the reaction mixture and the enzyme reaction was continuously monitored by measuring the change in absorbance at 475 nm of formation of the DOPA chrome for 1 min. |
