| Description: | MDCK-MDR1 or MDCK-MRP1 cells (50,000 cells per well) were seeded into black CulturePlate (96-well plate) with 100 μL medium and allowed to become confluentovernight. Hundred microliters of test compounds was solubilized in culture medium and added to monolayers. 96/wells plate was incubated at 37 °C for 30 min. Calcein-AM was added in 100 μL of phosphate-buffered saline (PBS) to yield a final concentration of 2.5 μm, and plate was incubated for 30 min. Each well was washed three times with ice-cold PBS. Saline buffer was added to each well, and the plate was read to Victor3 (PerkinElmer, Waltham, MA, USA) at excitation and emission wavelengths of 485 and 535 nm, respectively. |
