Assay Method Information

Assay Name:  Carbonic Anhydrase Inhibition Assay
Description:  The in vitro inhibition of carbonic anhydrase was assessed by a colorimetric assay. Carbonic anhydrase-catalysed hydrolysis of p-nitrophenyl acetate produces p-nitrophenol, which has an absorption peak at 384nm, which allows for colorimetric determination of product and subsequent calculation of enzyme activity. The percentage inhibition was defined relative to the maximal activity, which was measured without inhibitor. IC50 values were obtained from a graph of percent inhibition versus inhibitor concentration and calculated using Prism (Graphpad Software, CA).
