| Description: | The phosphatase activity resulted in the formation of the colored product p-nitrophenol, which was continuously monitored at 405 nm every 30 s for 15 min using a plate reader. For each concentration of test compound or DMSO control, the initial rates were used to fit the rectangular hyperbola of Michaelis-Menten by nonlinear regression analysis. The ratio of the apparent Km/Vmax vs inhibitor concentration was plotted, and the Ki was calculated by linear regression to be the negative X-intercept. |
