| Description: | The reaction was initiated by the addition of 150pmoles of enzyme to a mixture of inhibitor (40µM, 20 µM, 10 µM, 5 µM, 1 µM and 0.5 µM ), 20µM chorismic acid in 50mM Tris-HCl, 0.5mM EDTA, 0.1mg/ml BSA, 10mM β-mercaptoethanol (pH-7.5). The assay mixture was incubated at 37°C for 5 min and the reaction was terminated by the addition of 40µL of 1N HCl and further incubation at 37°C for 1 to 5 min. Phenyl pyruvate formed at the end of reaction is converted to chromophoric base with the addition of 80µL 2.5N NaOH. The absorbance of each sample was read at 320nm. Positive control contained the assay mixture with carvacrol(40µM, 20 µM, 10 µM, 5 µM, 1 µM and 0.5 µM ). |
