| Description: | IC50 values against APN from Porcine Kidney were determined by using L-Leu-p-nitroanilide as a substrate. All the solutions of the inhibitors were prepared in the assay buffer( pH 7.5). All the inhibitors were preincubated with APN for 5 min at room temperature. The assay mixture, which contained the inhibitor solution (concentration dependent on the inhibitor), the enzyme solution (4 mg/mL final concentration), and the assay buffer, was adjusted to 200 μL, and then incubated at 37 ℃ for 30 min. The hydrolysis of the substrate was monitored by following the change in the absorbance measured at 405 nm with the UV-vis spectrophotometer Pharmacia LKB, Biochrom 4060. |
