| Description: | Kinase assay: After the buffer was prepared, the enzyme was mixed with different concentrations of pre-diluted compounds in duplex, and allowed to stand at room temperature for 30 minutes. The corresponding substrate and ATP were added and reacted at room temperature for 60 minutes (in which negative and positive controls were set). After the reaction was completed, an antibody was added for detection. After incubation for 60 minutes at room temperature, detection was carried out with Evnvision and data were collected. The enzyme activity in the presence of each concentration of the compounds disclosed herein was determined by Evnvision microplate reader, and the inhibitory activity of the compounds at different concentrations on the enzyme activity was calculated. Then, according to the four-parameter equation, the inhibitory activity of the compounds at different concentrations on the enzyme activity was fitted using Graphpad 5.0 software, and the IC50 value was calculated. |
