| OGG1 Inhibitor Assays |
Description: | In view of the deficiencies with conventional assays and inhibitors, many embodiments are directed to assays that directly measure excision by OGG1. In various embodiments, the assay incorporates a fluorogenic probe (OGR1) configured to directly measure the base excision activity of OGG1 by detecting the excision of 8-OG in real-time (FIG. 1). (See, e.g., Edwards, S. K., et al., ChemBioChem 2015, 16, 1637-1646, the disclosure of which is incorporate herein by reference.) In embodiments of this probe, 8-OG acts as a fluorescence quencher of a neighboring fluorescent DNA base, and enzymatic excision of the 8-OG renders the probe emissive. In various other embodiments, fluorescence intensity increases may be used to supply quantitative assays of OGG1 activity. |
