| Description: | SERT assay (Compounds of Formula I): Test compounds (conc range), reference compound or vehicle control were incubated for 180 min at room temperature with CHO cells stably transfected with the human serotonin transporter (5×103 cells/well) with 0.15 μM [3H] serotonin in the presence or absence of the test or reference compound in buffer containing (in mM); Tris/HCl (pH 7.4) (5), HEPES/Tris (7.5), NaCl (120), KCl (5.4), CaCl2 (1.2), MgSO4 (1.2), Glucose (5) and ascorbic acid (1). Following incubation, [3H] serotonin uptake was quantified by a standard scintillation counting method. |
