| | Thioredoxin Reductase 1 (TrxR1) Inhibitory Activity Assay |
| Description: | Thioredoxin is known to influence melanogenesis substrates [Y Lu et al., Modulating skin colour: role of thioredoxin and glutathione systems in regulating melanogenesis, Bioscience Reports, 2021; 41 BSR20210427]. Accordingly, a study was undertaken to assess the impact, if any, of zinc di-(di-n-butyryl lysinate) (ZDBL), Ca di-(di-n-butyryl lysinate) and combinations thereof on the inhibition of thioredoxin reductase. Test materials were prepared by dissolving 0.01 gm of each test material in 1 ml ethanol to make stock solutions. For the analysis, 1 to 4 serial dilutions of the stock solutions using buffer (100 mM Potassium phosphate pH 7.0 containing 10 nM EDTA) were prepared to determine the IC50 values. Thioredoxin reductase 1 (Raybiotech, Catalog #228-12536-2) was added to the sample and the mixture was incubated for 1 hour at room temperature. After incubation, the working buffer (Phosphate buffer containing 2 mM DTNB and 0.2 mM NADPH) was added. The plate was read at 412 nm for 30 minutes to determine the inhibition. |
