The highest concentration of the compound detected was 1000 nM, 3-fold dilution, a total of 11 concentrations (1000 nM-0.017 nM). The reaction system is 10 nM FL MALT1 protein, 200 mM Ac-LRSR-AMC, 50 mM Tris pH 7.5, 0.6M Citrate, 1 mM DTT, 1 mM EDTA, 0.05% BSA and 1.5% DMSO. The compound and the enzyme were pre-incubated at room temperature for 50 min. The substrate was then added and the reaction was carried out for 4 h. |
