| Description: | Enzyme activity was assayed at 37° C. in 50 μL reaction mixtures containing 100 nM IP6K1 or IP6K2, or 200 nM IP6K3, and unless otherwise indicated, 5.0 μM human Dipp1,45 20 mM HEPES (pH 7.2), 100 mM KCl, 3.5 mM MgCl2, 20 μM EDTA, 25 μM InsP6 and 500 μM ATP for 60-120 min. Pi release was determined with a malachite green colorimetric assay.45 Where indicated, TNP (Cayman Chemical) was added to the assays. |
