| Description: | The enzymatic assay was carried out at room temperature in 96-well plates. The initial rate of PTPase-catalyzed hydrolysis of p-nitrophenol phosphate (pNPP) was measured by following the absorbance change at 405 nm. IC50 value was determined under fixed pNPP concentration of 1 mM. All the assays were carried out in duplicate or triplicate and the average results are presented. Ki is derived from IC50 based on competitive inhibition Ki = IC50 x Km/ (Km + [substrate]). |
