| Description: | To determine the inhibitory potency of the drugs, the initial velocity of the DHODR-catalyzed reaction at saturating concentrations of DHO (1 mM) and QD (0.1 mM) was measured, with varying drug concentrations. The equation vi /vo= 1/[1 + [I]/IC50], where vi is the initial velocity in the presence of the inhibitor at concentration [I] and vo is the initial velocity in the absence of the inhibitor, was fitted to the initial velocities in order to find the drug concentration causing 50% inhibition of the enzyme activity. |
