| Description: | Inhibition assays were performed in duplicate using two different concentrations of human IMPDH type 1 (87 and 155 nM) and varying concentrations of inhibitor. The production of NADH was monitored by following changes in absorbance at 340 nm on a Hitachi U-2000 spectrophotometer. Steady-state velocities were used to determine Ki(app) values by fitting the velocities vs inhibitor concentration to a simple binding model with Dynafit. |
