Assay Method Information

Assay Name:  CHK1 AlphaScreen
Description:  AlphaScreen (PerkinElmer Life Sciences, Inc) was performed in opaque 384-well polypropylene plates. The assay relies on hydrogel coated Donor and Acceptor beads providing functional groups for conjugation to biomolecules. Binding of molecules captured on the beads leads to an energy transfer from one bead to the other, ultimately producing a luminescent/fluorescent signal. Streptavidin-coated donor beads and protein A-conjugated acceptor beads were added together. After incubation for > 6 hours in the dark at room temperature, the assay end point was measured using a Fusion Multilabel Reader, 300 ms excitation at 680 nm and 700 ms per well reading time at 520-620 nm. The signal was expressed in counts per second. Primary screening data were analysed in Excel (Microsoft). Percentage inhibition was calculated.
