| Description: | The assay was carried out in 384-well plates according to literature procedures [46]. Each well contained 100mM sodium phosphate and 0.2mM EDTA pH 7.0. 10ng PDI was pre-incubated with various concentration of probes (4% DMSO) in 20uL buffer for 30 min at 37 degrees Celsius, followed by the addition of insulin (0.16mM final concentration) and DDT (1mM final concentration). The enzyme reaction was monitored at 650nm on a Bioteck microplate reader. |
