| Description: | One nanomolar of proteasome was incubated with different inhibitor concentrations in Tris 25 mM pH 7.5; SDS 0.03%; EDTA 0.5 mM for 15 min at 37 °C in 96-well plates, in a final reaction volume of 40 mL. Ten microliters of specific fluorogenic proteasome substrate Suc-LLVY-Amc (CTL substrate)at 200 mM were added, resulting in a final substrate concentration of 40 mM. Samples were incubated for 15 min at 37 °C and then the fluorescence in each well was measured. |
