| Description: | The fluorescence anisotropy assay was performed with fluorescein-labeled SAHA (fl-SAHA, gift of C. Fierke, University of Michigan) and measured using a TECAN infinite F200Pro plate reader (λex = 485 nm, λem = 535 nm)60. Briefly, 30 nM fl-SAHA was titratedwith enzyme (0–15 μM) and incubated at room temperature for 20 min.The Kd values were determined by fitting the binding isotherm using GraphPad Prism software. |
