Assay Method Information

Assay Name:  functional assay
Description:  Nematode eggs are isolated from fresh feces by centrifugation and flotation. The eggs are washed and inspected to confirm embryonation has not started. Each sample is tested at six concentrations of a test compound (e.g., 0.02, 0.05, 0.1, 0.25, 0.5 and 1.0 μg/ml) and a negative control (no compound). After incubation for 48 h, the assay is ended, and all eggs and larvae present in each well are counted. The Hill equation with a variable slope is used to fit the dose-response relations using GraphPad Prism. The EC50-values and 95% confidence intervals are calculated.
