| Description: | Test compounds (conc range), reference compound or vehicle control were added to assay buffer containing (in mM); Tris/HCl, pH 7.4 (40), MgCl2 (8) and EGTA/NaOH (1.7), containing 450 nM cAMP (450 nM) and 0.25 μCi (PDE-4B1) or 0.0125 μCi (PDE-4D2) [3H] cAMP. 20 mins after addition of human recombinant PDE-4B1 (1.2 IEU) or PDE-4D2 (1.5 IEU), SPA beads were added and incubated at 22° C. for a further 30 mins. [3H]5′AMP was quantified by a standard scintillation counting method. |
