| Description: | Mammalian target of rapamycin (mTOR) was assayed by monitoring phosphorylation of GFP-4EBP using a homogeneous time-resolved fluorescence resonance energy transfer format. The mTOR-mediated phosphorylation was measured under initial rate conditions. After incubation, the phosphorylated substrate was detected with Tb-anti-p4E-BP1 antibody before reading on a Perkin-Elmer EnVision Fluorescence Reader (exc 340; em 495/520). Duplicate dose-response curves were fit to an equation of competitive tight-binding inhibition. |
