| Description: | The activity of ALR2 enzyme was determined at 340 nm in UV spectrophotometer that depends upon the measurement of NADPH consumption. Each well of the 96-well plate contains 200 無 of assay mixture consisting of potassium phosphate buffer 100 mMat 6.2 pH, 20 無 of 10 mM test sample, 70 無 of enzyme preparation and substrate D,L-glyceraldehyde (40 無), all mixture was incubated at 37 蚓 for 5 min and then 0.1 mM NADPH (50 無) was added as a cofactor in the enzyme reaction and read was measured at 340 nm and followed by incubation at 37 蚓 for 10 min and thenabsorbance was measured in Eliza plate reader at respective wavelength. Negative and positive control used were 10 m M buffer (20 無) and 10 mM sulindac (20 無) [Med. Chem. Commun., 5:1371-1380]. |
