| Description: | The PDK2 or Hsp90 N-terminal domain protein was dialyzed against 1 liter of thedialysis buffer containing 50 mM Tris-Cl, pH 7.5, 50 mM KCl, 1 mM MgCl2, and 0.5 mM β-mercaptoethanol. Known or novel PDK inhibitor solutions (150-1500 μM) were placed in the titration syringe and injected in 8-μl increments into the reactioncell containing 1.4 ml of 18-70 μM PDK2 or Hsp90 N-terminal domain at 15 °C in a VP-ITC microcalorimeter (GE Healthcare). |
