| Description: | ATR enzyme (batch: Eurofins Cat. No. 14-953) solutions were prepared in 1x kinase base buffer. 10 ul of 2x enzyme solution (final concentration: 2.5 nM) was added to each well of the 384-well assay plate containing 60 nl compound in each well. The plate was incubated at room temperature for 10 minutes. 2x peptide solutions were prepared with FAM-labeled peptide and ATP in the 1x kinase base buffer. 10 ul of 2x peptide solution was added to each well of the 384-well assay plate, which was incubated at 28 C. for 240 min. 40 ul of stop buffer was added to stop reaction. Data were collected by Caliper. |
